I don't mind the idea of creating 5 awesome heroes but i have 2 questions:
1. How long do we have? and
2. could i have a little more detail on how these designs are judged? eg. when you say:
"Up to 10 points can be given per skill, so you get up to 40 here. These 10 points include originality and simplicity."
do you mean simplicity as in balanced or destructive?

And by the way your "Amazon Queen's" second abilitity is ridiculous, if you consider timing and what's currently out there you would be blocking quite a few ult's and abilities in quite quick succession due to you "3" charges. Think about it, it's overpowered in the way that it could block the greatest of damage very easily. My suggestion would to either; make it one charge total with a 10 second cd or instead of total invunerability make it heavy armour bonus.

I've made all 5 of my heroes, but quite a few of their skills are still up in the air and I'm not sure what literary character one of them is yet. Time will tell.

If you look at my "point system", you'll realize that those concerns are completely unsubstantiated. Only 1/12 of the total points come from the overall theme.

Yeah, made up that name on the fly too. Anyways, I always loved simplicity over complexity. I feel uniqueness is defined by the full set of skills, not by how many effects you can cram into four slots or some fancy-pants mechanism the entire hero is built around.
