Every cinematic Valve has ever made was made with the Source Filmmaker. There's a lot of people in the company with a lot of experience using the software, so it would make sense that they use it to create the DotA 2 trailer. The quality in the trailer is lower than today's standard CGI as well, and matches the style of previous Valve cinematics for TF2 / L4D. It would be pretty stupid for them to outsource to some other CGI department when they can make the whole thing with their own people and their own software.
I'm not claiming it was definitely made with it, but it certainly makes sense that they would do it.

The only good thing about that horrible trailer is the voice acting.
