Look like i was mistaken the condition to make the rune earn a purple diamond (exp indicator).
All i it seems to need , is to be near enemy creeps (or i guest anything that would give you a purple diamond)
Confirmed, you just need to be in exp range of a unit, don't even need to be in range on the rune.
